اثر هشت هفته مصرف مکمل ترکیبی همراه با تمرین هوازی بر بیان ژن‌های ARC و IGF-1 هیپوکامپ و عملکرد شناختی در موش‌های مبتلا به آلزایمر: یک مطالعه تجربی

چکیده
مقدمه: ARC و Insulin-like growth factor 1 (IGF-1) از جمله نشانگرهای مولکولی مهم برای نروژنز مغزی هستند که در شرایط مختلف همچون آلزایمر، در بافت مغز به طور قابل توجهی کاهش می‌یابند. هدف از انجام پژوهش حاضر، بررسی تأثیر هشت هفته مصرف مکمل ترکیبی همراه با تمرین هوازی، بر بیان ژنی ARC و IGF-1 هیپوکامپ و عملکرد شناختی در موش‌های سالمند مبتلا به آلزایمر بود.
مواد و روش‌ها: در این مطالعه تجربی، 25 سر موش نر c57 مبتلا به آلزایمر با سن 16-14 هفته و وزن 35-30 گرم به طور تصادفی در گروه‌های «شاهد سالم، آلزایمر، آلزایمر + تمرین هوازی، آلزایمر + مکمل ترکیبی، آلزایمر + تمرین هوازی + مکمل ترکیبی» قرار گرفتند. القای آلزایمر با تزریق آمیلوئید بتا (Amyloid-beta یا Aβ1-42) به هیپوکامپ انجام شد. پروتکل تمرین هوازی به مدت پنج روز در هفته هشت هفته قبل و پس از القای آلزایمر بود. مکمل ترکیبی شامل رزوراترول با غلظت 25 میلی‌گرم بر کیلوگرم و فیستین با غلظت 20 میلی‌گرم بر کیلوگرم به مدت هشت هفته استفاده گردید. داده‌ها با استفاده از آزمون‌های One-way ANOVA و تعقیبی Tukey در سطح معنی‌داری 05/0 مورد تجزیه و تحلیل قرار گرفت.
یافته‌ها: القای آلزایمر باعث کاهش معنی‌دار بیان ژن‌های ARC و IGF-1 (001/0 = P)، افزایش معنی‌دار بیان ژن Amyloid precursor protein (APP) در هیپوکامپ (001/0 = P) و کاهش معنی‌دار عملکرد شناختی هیپوکامپ (001/0 = P) شد. تمرین هوازی و مصرف مکمل ترکیبی هم به صورت جداگانه و هم به صورت هم‌زمان، باعث بهبود معنی‌دار تمام تغییرات فوق گردید (001/0 = P).
نتیجه‌گیری: به نظر می‌رسد تمرین هوازی و مصرف مکمل ترکیبی، احتمالاً می‌تواند از طریق افزایش بیان ژنی ARC و IGF-1 در هیپوکامپ، به افزایش نروژنز مغزی و بهبود عملکرد شناختی در افراد سالمند مبتلا به آلزایمر کمک نماید.
