<ArticleTitle>The Attenuating Effect of High-Intensity Interval Training and Nicotinamide Mononucleotide Supplementation on Aging-Related Oxidative Stress in Rat&#039;s Heart Tissue</ArticleTitle>

		<Abstract>Introduction: Numerous studies have shown attenuating effects of nicotinamide mononucleotide (NMN) and high-intensity interval exercise (HIIT) training on reducing oxidative stress. Therefore, in the present study, we evaluated the effect of HIIT and NMN supplementation on the oxidative stress markers in cardiac tissue of rats.Materials and Methods: 48 sprago dauli rats with an average age of 8 to 10 months were randomly divided into 6 groups of pretest control, control, sham (normal saline), HIIT, NMN supplementation, and combination (HIIT + NMN). The HIIT and combination groups performed HIIT on the treadmill for 8 weeks, 3 sessions per week. NMN with a dose of 500 mg per kilogram body weight was administered intra-peritoneally in the NMN and combination groups. 24 hours after the last training and NMN administration, the animals were sacrificed, and their hearts were removed for evaluating oxidative stress factors [malondialdehyde (MDA), protein carbonyl (PC), superoxide dismutase (SOD), and glutathione peroxidase (GPx)]. One-way ANOVA test was used for statistical analysis.Results: The expression of GPx gene in the groups receiving NMN, HIIT, and combination was significantly higher compared to the pretest control, control, and sham groups (P &lt; 0.050). Moreover, the levels of MDA and PC were lower in NMN, HIIT, and combination groups compared to the pre-test control, control, and sham (P &lt; 0.050). The highest reduction was seen in the combination group (P &lt; 0.001).Conclusion: It seems that regular consumption of NMN and/or HIIT may attenuate oxidative stress in the cardiac tissue; however, combinations of NMN and HIIT significantly improves the effects of sole interventions.</Abstract>
			<OtherAbstract Language="FA">مقدمه: هرچند نقش تمرینات ورزشی و مکمل‌دهی نیکوتین‌آمید مونونوکلئوتید (Nicotinamide mononucleotide یا NMN) بر بهبود استرس اکسیداتیو تأیید شده، اما اثر تعاملی این دو متغیر در بافت قلب هنوز به طور کامل شناخته نشده است. پژوهش حاضر با هدف بررسی اثر تمرینات تناوبی شدید (High intensity interval training یا HIIT) و مصرف مکمل NMN بر استرس اکسیداتیو در بافت قلب موش‌های صحرایی انجام شد.مواد و روش‌ها: 48 رت نژاد اسپراگو داولی 8 تا 10 ماهه، به صورت تصادفی در شش گروه شاهد هفته اول (شاهد پیش از مطالعه)، شاهد هفته آخر (شاهد)، دارونما (دریافت نرمال سالین)، تمرین HIIT، مکمل NMN و ترکیبی (NMN + HIIT) تقسیم شدند. گروه‌های تمرین و ترکیبی سه جلسه در هفته به مدت 8 هفته، HIIT را روی تردمیل انجام دادند. 500 میلی‌گرم مکمل NMN نیز به ازای هر کیلوگرم وزن بدن به صورت داخل صفاقی برای گروه‌های NMN و ترکیبی تجویز گردید. 24 ساعت پس از آخرین تمرین و تجویز NMN، حیوانات قربانی شدند و قلب آن‌ها جهت ارزیابی شاخص‌های استرس اکسیداتیو [مالون دی‌آلدئید (Malondialdehyde یا MDA)، پروتئین کربونیل (Protein carbonyl یا PC)، بیان ژن‌های سوپراکسید دیسموتاز (Superoxide dismutase یا SOD) و گلوتاتیون پراکسیداز (Glutathione peroxidase یا GPX)] خارج گردید. داده‌ها با استفاده از آزمون One-way ANOVA مورد تجزیه و تحلیل قرار گرفت.یافته‌ها: بیان ژن GPX در گروه‌های NMN، HIIT و ترکیبی در مقایسه با گروه‌های شاهد و شم به طور معنی‌داری افزایش یافت، اما این افزایش در گروه ترکیبی نسبت به بقیه گروه‌ها بیشتر بود (050/0 &gt; P). همچنین، میزان MDA و PC در این گروه‌ها نسبت به گروه‌های شاهد و شم کاهش یافت که بیشترین کاهش در گروه ترکیبی مشاهده شد (001/0 &gt; P).نتیجه‌گیری: به نظر می‌رسد مصرف NMN و انجام HIIT می‌تواند استرس اکسیداتیو را در بافت قلب کاهش دهد، اما ترکیب NMN و HIIT تأثیرات بیشتری دارد. بنابراین، می‌توان با ترکیب HIIT و مکملNMN، به کاهش بیشتر استرس اکسیداتیو و افزایش آنتی‌اکسیدان دست یافت.</OtherAbstract>
